The first specific small molecule inhibitors of translation termination i bacteria

Virtual screening of a large library of small drug-like molecules (3.4 M) against the 3D structures of bacterial release factors delivered a number of potential hits. These were then tested experimentally both for their ability to inhibit bacterial growth and block translation termination in vitro. Two of the compounds were found to inhibit termination in cell-free fast kinetics experiments and were also found to be bactericidal.
